چکیده
Seaweeds emerge as promising third-generation renewable for sustainable bioproduction. In the present work, we valorized brown seaweed to produce l -lysine, world's leading feed amino acid, using Corynebacterium glutamicum , which was streamlined by systems metabolic engineering. The mutant C. SEA-1 served a starting point development because it produced small amounts of -lysine from mannitol, major sugar, deletion its arabitol repressor AtlR and engineered pathway. Starting SEA-1, systematically optimized microbe redirect excess NADH, formed on sugar alcohol, towards NADPH, required synthesis. mannitol dehydrogenase variant MtlD D75A, inspired 3D protein homology modelling, partly generated NADPH during oxidation fructose, 70% increased yield in strain SEA-2C. Several rounds engineering further supply production. best strain, SEA-7, overexpressed membrane-bound transhydrogenase pntAB together with codon-optimized gapN encoding NADPH-dependent glyceraldehyde 3-phosphate dehydrogenase, mak fructokinase. fed-batch process, SEA-7 76 g L ?1 at 0.26 mol maximum productivity 2.1 h . Finally, integrated into valorization cascades. Aqua-cultured Laminaria digitata commercial alginate, extracted hydrolyzed enzymatically, followed recovery clean-up pure alginate gum. residual sugar-based mixture converted 0.27 C-mol SEA-7. Second, stems wild-harvested Durvillaea antarctica obtained waste processing blades human consumption, were acid treatment. Metabolic engineering of Corynebacterium glutamicum for L-serine production.
Although L-serine proceeds in just three steps from the glycolytic intermediate 3-phosphoglycerate, and as much as 8% of the carbon assimilated from glucose is directed via L-serine formation, previous attempts to obtain a strain producing L-serine from glucose have not been successful. We functionally identified the genes serC and serB from Corynebacterium glutamicum, coding for phosphoserine ...
متن کاملEngineering Corynebacterium glutamicum to produce 5-aminolevulinic acid from glucose
BACKGROUND Corynebacterium glutamicum is generally regarded as a safe microorganism and is used to produce many biochemicals, including L-glutamate. 5-Aminolevulinic acid (ALA) is an L-glutamate derived non-protein amino acid, and is widely applied in fields such as medicine and agriculture. RESULTS The products of the gltX, hemA, and hemL genes participate in the synthesis of ALA from L-glut...
متن کاملMetabolic flux engineering of L-lysine production in Corynebacterium glutamicum--over expression and modification of G6P dehydrogenase.
In the present work, metabolic flux engineering of Corynebacterium glutamicum was carried out to increase lysine production. The strategy focused on engineering of the pentose phosphate pathway (PPP) flux by different genetic modifications. Over expression of the zwf gene, encoding G6P dehydrogenase, in the feedback-deregulated lysine-producing strain C. glutamicum ATCC 13032 lysC(fbr) resulted...
متن کاملFermentative Production of Lysine by Corynebacterium glutamicum from Different Carbon Sources
Production of lysine by Corynebacterium glutamicum (PTCC 1532) from different agricultural by-products (molasses and pulpy waste date) was compared to glucose as raw materials. For this purpose, ammonium sulphate was selected as a constant nitrogen source. The effect of different nitrogen sources was also investigated with glucose as a constant carbon source. The production of L-lysine was exam...
